The incumbent contributes to programs that apply molecular biology and genetics to the modification of microorganisms and cell lines that express vaccine antigens of interest. Subject antigens may be proteins expressed in E. coli, mammalian cells or other appropriate host backgrounds or antigens expressed in the native host, such as polysaccharides. Studies directed toward improving the growth and productivity of bacterial antigen production strain is the major responsibility of the position. Experiments that apply diverse genetic techniques in a variety of host backgrounds to improve product yield or quality is also in scope. A major component of the position is devising techniques to efficiently prepare cell banks for new organisms/strains that could be readily applied in a cGMP environment. Experience in microbiology and genetic is required. An appreciation of cell banking strategies would be helpful
Performs experiments that identify optimized conditions for growth and antigen expression in bacterial strains of interest.
Performs experiments that identify optimized conditions for preservation of bacterial strains of interest.
The incumbent will be responsible for performing experiments that create new cell banking and testing strategies for both laboratory strains and those of commercial interest
Performs laboratory experiments that evaluate strains, media and expression strategies for production of vaccine antigen candidates
The incumbent will assist in experiments directed to preparing targeted mutants of E. coli for use as specialized hosts to support recombinant protein expression in fermentation
The incumbent will assist in the construction of recombinant plasmids
The incumbent will be responsible for the preparation of bulk plasmid preparations for use in mammalian transfection experiments and in vitro transcription
Incumbent assists senior researchers in the evaluation of new technologies for the group
Incumbent evaluates and applies new analytical methods for the group
Gains understanding of the discipline through reading of the scientific literature
Organizes and tabulates data
Presents data summaries within the group
Fully documents experimental results in notebooks in a timely and compliant manner.
Works irregular hours (weekends and outside of 9-5 format) as experiments warrant
MS/BS degree in microbiology, biotechnology, bioengineering or microbial genetics with 3-5 years of relevant experience
Has a solid understanding of microbiology and an appreciation of molecular biology and/or microbial genetics
Has previously worked in a mirobiology, biotechnology or microbial genetics laboratory setting
Experience in formal microbial cell banking and familiarity with testing protocols and guidelines is preferred
Experience in the genetic manipulation of E. coli and/or other industrially important gene expression hosts is preferred
Has knowledge of cGMP and regulatory requirements as they apply to preparation of cGMP cell banks
Has experience with analytical techniques such as SDS-PAGE and HPLC
Ability to organize and execute experiments
Ability to work with limited supervision
Collaborates in a team environment
Has a proven ability to organize and tabulate data
