This position offers the opportunity to join the newly established Single Cell Genomics Core at Brigham and Women’s Hospital. Located in the Brigham and Women’s Hospital Human Immunology Center, the Core has been established to perform single cell gene expression profiling through the 10X Genomics pipeline. U nder general supervision by a supervisor, and in accordance with established hospital policies and procedures, the self-motivated individual will perform activities related to the operation of the Core, including processing samples and operating the 10X Genomics machine, performing library preparation reactions, and interacting with research scientists. Responsibilities also include managing scheduling and billing for the core, ordering reagents and supplies, upkeep and quality control of the machines, and interactions with team members and other Cores. In addition, this individual will perform a variety of routine assays and experiments in the laboratory and help in general to maintain lab organization and support laboratory staff.
1. Operate the 10X Genomics instrument and downstream processing:
Interact with research scientists to prepare samples for 10X Genomics analyses
Process samples through the 10X machine
Perform routine maintenance and quality control assessments of the instrument
Perform sample library preparation and preparation of sample for sequencing
Coordinate sequencing of samples with RNAseq Core
Perform duties required for daily operation of the Core
Monitor and order reagents
Manage scheduling and track utilization of the Core
Interact with researchers to answer questions and provide guidance
Demonstrate basic use of RNAseq analysis software to users
Process samples for research, including:
a. Processing of tissue samples and isolation of cells
b. Cell staining for flow cytometry and cell sorting
c. Isolation and analysis of RNA
d. Aliquot and freeze or thaw biospecimens
e. Limited operation of the flow cytometry cell sorter
Work independently under general supervision, perform a variety of routine assays and experiments in the laboratory including (but not limited to):
Short-term and long-term cell cultures requiring aseptic cell culture technique
Functional in vitro cell culture assays
5. Report on progress of work at laboratory seminars.
6. Prepare laboratory reagents, chemicals, instruments and equipment.
7. Maintain supplies inventory in accordance with laboratory and department procedures.
8. Assist in the orientation and training of new students, technicians, and fellows.
9. Maintain established department policies, procedures and objectives, including the areas of quality assurance, safety, environmental and infection control.
10. Perform other duties as directed.
ü Bachelor’s degree or equivalent required.
ü At least one year experience in a biomedical research lab with molecular biology experience.
ü Demonstrated organizational and interpersonal skills as required to work closely with a research team.
ü Demonstrated competence with databases/spreadsheets and software packages.
ü Willingness to learn other laboratory techniques.
SKILLS/ ABILITIES/ COMPETENCIES REQUIRED: (MUST be realistic, neither overstated nor understated, and related to the essential functions of the job.)
ü Must possess good organizational skills.
ü Ability to communicate information clearly in both oral and written form, including email.
ü Ability to make decisions based on logical assumptions of factual data to solve technical and logistical problems.
ü Experience with flow cytometry preferred.
