Spectrix is seeking a highly motivated Scientist to join our client's bioconjugation team within DBTx to contribute to the development of antibody-drug conjugates (ADCs) to support the oncology and PH ADC portfolio. This offers an excellent opportunity to gain hands-on experience in conjugation of a variety of different linker-payloads to antibodies, inlcuding cytotoxic and degrader payloads. The individual(s) will perform conjugation, purification and analytics of ADCs, and will provide critical contributions to the expanding bioconjugation portfolio.
Roles and Responsibilities
- Design, produce, and characterize antibody-drug conjugates (ADCs) on a variety of scales, and in alignment with product development guidelines and QC criteria, including associated purification and analytical characterization of the conjugates
- Develop and optimize methods for performing plate-based bioconjugations for the production of ADCs
- Maintain thorough, accurate, and timely experimental records in the Electronic Laboratory Notebook (ELN), and register conjugates and upload supporting analytical data to the appropriate sample-tracking and data-management systems
Experience, Education and Specialized Knowledge and Skills
· Associates or Vocational Degree + 3-5 years of academic / industry experience or Bachelor's Degree + 1-3 years of academic / industry experience or Master's Degree + 0-1 years of academic / industry experience
Required skills:
· Strong background in biochemistry, protein sciences, bioconjugation, analytics, organic chemistry and/or chemical biology
- Hands-on experience in the discovery and development of protein-based therapeutics, preferably antibodies and/or ADCs. Experience in optimizing and performing protein purification, including various chromatographic techniques such as desalting, affinity, size exclusion, ion-exchange, and hydrophobic interaction chromatography
- Demonstrated experience in analytical characterization and QC of proteins/antibodies, including AnSEC, RP-HPLC, LC-MS, CE-SDS, ELISA, DSC, and HIC
- Demonstrated ability to work effectively within a team environment
- Excellent organizational, analytical and technical skill, along with demonstrated initiative-taking and innovative thinking
- Enthusiasm, willingness to learn, strong work ethic, broad scientific interests
- Excellent interpersonal and communication skills along with a sense of urgency and ability to work on multiple projects simultaneously
Preferred skills:
- Working knowledge of common bioconjugation chemistries and workflows, including lysine (amine) conjugation, reduced inter-chain cysteine conjugation, and site-specific/engineered cysteine conjugation.
- Familiar with common protein engineering strategies for conjugation, such as engineered cysteines or the incorporation of non-canonical amino acids for site-specific attachment
- Experience with enzymatic conjugation methods (e.g., microbial transglutaminase-, glycan remodeling, or sortase-mediated conjugation)
- Experience working with plate-based assays or performing plate-based conjugations
