The Department of Biological Sciences in the College of Arts and Sciences at Lehigh University invites applications for a Pre-Doctoral Research Associate (Lab Technician) to join the laboratory of Professor Damian Dudka. The Dudka Lab (dudkalab.me) investigates how the rapid evolution of centromeric DNA and centromere-associated proteins regulates mammalian chromosome segregation and the impacts on reproduction, development, and human disease. This is an excellent opportunity for a recent graduate seeking substantive bench experience in mammalian cell and reproductive biology prior to a graduate or medical program.
About the Role:
The Pre-doctoral Research Associate will support the day-to-day operation of a new evolutionary cell biology laboratory and contribute to ongoing research projects in the lab. Core duties span molecular biology, tissue culture, confocal microscopy, mouse colony management, and general lab management. The successful candidate will work closely with the PI and gain extensive hands-on training in techniques used in modern cell, reproductive, and evolutionary biology research, with opportunities to contribute to peer-reviewed publications.
Key Responsibilities:
- Maintain mammalian cell culture lines (mouse embryonic stem cells, mouse/human cell lines), including media preparation, passaging, and freezing/thawing.
- Perform standard molecular biology techniques: plasmid design, RNA/DNA isolation, PCR/qPCR, bacterial transformation, and plasmid isolation.
- Assist with the mouse colony, including breeding, genotyping (DNA extraction and PCR), weaning, and coordination with the vivarium under approved IACUC protocols.
- Support oocyte and preimplantation embryo experiments, including dissection, in vitro culture, immunofluorescence preparation, and assistance with microinjection workflows.
- Acquire and analyze data on confocal microscopes (with training), image quantification.
- Manage general lab operations: ordering and inventory of reagents and consumables, equipment maintenance and service coordination, biosafety, and chemical waste compliance, and lab safety documentation.
- Train and assist undergraduate and rotation students with routine techniques under the PI’s supervision.
- Maintain detailed, organized electronic and paper laboratory records.
Qualifications:
- Bachelor's degree (or near completion) in biology, biomedical sciences, molecular biology, genetics, or a closely related field by the start of the appointment.
- Prior research experience (undergraduate research, senior thesis, summer internship, or equivalent) in a wet-lab setting.
- Familiarity with at least some of the following: mammalian cell culture, basic molecular cloning, PCR, gel electrophoresis, and immunofluorescence techniques.
- Strong organizational skills, attention to detail, and the ability to maintain accurate records.
- Strong written and verbal communication skills; ability to work both independently and as part of a collaborative research team.
- Commitment to fostering an inclusive research environment.
Preferred Qualifications:
- Experience with mouse handling, mouse colony management, and mouse genotyping.
- Experience with CRISPR-based gene editing
- Experience with confocal microscopy and image quantification (Fiji/ImageJ).
- Experience with biosafety procedures at the BSL-2 level.
Terms of Appointment:
This is a full-time (40 hours/week), one-year position with the possibility of renewal up to five years based on performance, research progress, and the availability of funds. The position is non-exempt and qualifies for all Lehigh University benefits, except that Research Associates receive an annual two-week leave of absence in lieu of vacation during the appointment period. Continued employment is contingent upon remaining in good standing with the University, making satisfactory progress on the research program, and the availability of funding. This is a full-time position with a 1-year initial appointment and the possibility of renewal based on performance and funding availability. The position will start on a mutually agreed-upon date.
