Full Job Description

Senior Scientist/Principal Scientist, Regulatory T-cell Immunology

Sift Bio is building a cutting-edge proprietary platform that combines AI, metagenomics, and high-throughput T-cell screening to discover epitopes that drastically amplify pre-existing immune memory responses, unlocking targeted, effective, and safe treatment for cancers and autoimmune diseases where current therapies fall short. Spinning out of UC Berkeley, the Sift Bio team brings together AI-driven discovery and deep translational immunology, with a proven track record of advancing novel immunotherapies from early concept through IND-enabling development. Collectively, the team has built and scaled high-throughput discovery platforms and led T-cell, peptide, and cell-based immunotherapy programs across leading academic labs, biotech startups, and biopharma. Merging this experience with proprietary datasets and computational pipelines, Sift Bio is shaping a new frontier in immunotherapy, unlocking the power of pre-existing immunity to create a fundamentally new class of treatments for immune dysfunctions. Our ultimate goal is to bring durable disease control to every patient.

We are seeking a Principal Scientist / Senior Scientist, Regulatory T-Cell Immunology to lead preclinical drug discovery, focusing on exploiting immunosuppressive T-cells to treat autoimmune diseases, cancer, and inflammation. This role will design and direct experiments, engineer novel cell therapies, and guide research strategy, manage project timelines, and collaborate with teams across clinical development, pharmacology, and regulatory affairs. This role will report to CTO and be fully onsite at 733 Industrial Road, San Carlos, CA.

KEY RESPONSIBILITIES:

Build, qualify, troubleshoot, and optimize end-to-end Treg suppression assays, including selection of potency readouts such as proliferation, cytokine production, activation markers, and other functional measures.
Create ex vivo assays that measure how antigen-specific T-cells drive suppression, benchmark findings against competing modalities, and diagnose underperforming Treg expansion programs through rigorous experimental design.
Characterize candidate epitopes using standard cellular, molecular, genetic, and immunological approaches, including antigen-specific Treg detection: tetramer/multimer staining and antigen-driven activation-induced marker (AIM) readouts, with matrix/pool deconvolution design for peptide screening.
Design and direct in vitro, ex vivo, and in vivo studies to evaluate mechanism of action, target biology, therapeutic efficacy, safety, persistence, stability, and suppressive function of candidate epitopes.
Develop and implement multi-parameter flow cytometry panels, functional immune assays, lineage-stability readouts, and disease-relevant models to support target validation, lead selection, and translational decision-making.
Lead external collaborations, CRO studies, and academic partnerships as needed, ensuring scientific rigor, timeline adherence, and data quality.

REQUIRED EXPERIENCE:

PhD in Immunology, Cell Biology, or a related field, plus 5+ years of post-doctoral or industry experience. Alternatively, a Master’s degree with 8 to 10+ years of biotech experience. Job title will commensurate with experience.
Excited by and scientifically driven to overcome complex technical challenges that will contribute to safe treatment for cancers and autoimmune diseases.
Deep hands-on experience with primary human Tregs and peripheral blood mononuclear cells (PBMCs), including working knowledge of best methods for isolation, expansion, and activation of Tregs/PBMCs
Prior experience developing antigen-specific tolerance or Treg-based approaches, peptide, protein, or engineered cell therapy setting, spanning antigen selection through in vivo functional readouts
Strong T-cell biology expertise, with extensive experience characterizing low frequency (<0.1%) CD8 and CD4 responses, and knowledge of how to improve readouts of rare T-cell responses
Knowledge of how to qualify Treg-based assays with known controls, for how to “rank” performance of manipulations -> impact Treg biology, and methods for how to overcome limitations of characterizing rare T-cell populations
Proven ability to contribute to the scientific direction of a project.
Strong written and oral communication skills for technical and non-technical audiences.
Conscientious team member with experience taking and giving constructive feedback.
Must be comfortable with executing lab work full time in a preclinical-stage startup environment, and capable of finding solutions to tough problems with limited resources.

PREFERRED QUALIFICATIONS:

Familiarity with HLA-transgenic mouse models and/or human PBMC xenograft/humanized models, and a clear-eyed view of which models are best suited to readout effects of an antigen-directed tolerance approach
Experience building immunotherapies for oncology or autoimmune disease settings
